●Key Characteristics: IGF-DES is a truncated analogue of the natural hormone insulin-like growth factor-1 (IGF-1). Specifically, it lacks the first three amino acids (N-terminal tripeptide: glycine-proline-glutamate) of the full-length IGF-1 molecule. This modification is not naturally present in the human body.
●Origin: It stems from research on the structure-activity relationship of IGF-1. Scientists discovered that removing the first three amino acids significantly alters the interaction between this peptide and its binding proteins.
●Main Difference from IGF-1: Full-length IGF-1, when circulating in the blood, primarily (over 99%) binds to the IGF-binding protein (IGFBP) family. This binding stabilizes IGF-1 but also severely limits its bioavailability and potency at the tissue level, acting as a storage and regulatory mechanism.
●"DES" Advantage (Theoretical): Deleting the first three amino acids significantly reduces the affinity of IGF-DES for most IGFBPs, especially the main carrier IGFBP-3. Theoretically, this would lead to:
○ A significantly increased free fraction: IGF-DES contains a much higher proportion of biologically active, unbound ("free") components compared to IGF-1.
○ Enhanced biological activity: It is believed that this increased free fraction, at molar concentrations, can significantly improve receptor activation potency in target tissues such as muscle and cartilage compared to IGF-1. Studies have shown that its potency may be 10–100 times that of IGF-1 in vitro and in some animal models.
