Product Introduction

Humanin is a 24-amino acid endogenous signal peptide encoded by mitochondrial DNA, belonging to the mitochondrial-derived peptide (MDP) family. It was initially discovered in screening experiments for Alzheimer's disease neuronal survival. It overturned conventional wisdom, demonstrating that mitochondria, in addition to encoding respiratory chain components, can also produce bioactive peptides capable of intracellular and extracellular communication. As a raw material for freeze-dried peptide research, it is not limited to a single tissue target, but can simultaneously protect intracellular mitochondria and facilitate extracellular receptor signal transduction. Its endogenous expression level gradually declines with age and is correlated with age-related cellular vulnerability.

Humanin's most fascinating advantage lies in its versatile, broad-spectrum protective effects. Early research brought it fame for its ability to protect the brain from Alzheimer's-related neurotoxicity, but subsequent scientific exploration has shown that its scope extends far beyond this. It not only improves overall insulin sensitivity and participates in the regulation of glucose metabolism, but also plays a crucial role in maintaining vascular function, resisting ischemic damage, and even slowing down the overall aging process. As scientists discovered that it belongs to a large family of "mitochondrial-derived peptides," the central role of humanin as a prototype molecule in metabolic health and longevity pathways is becoming increasingly clear.

Objective Shortcomings

Containing two methionines, Met¹ and Met²⁰, plus a Cys⁸ free sulfhydryl group, its activity is significantly reduced due to two main degradation pathways in aqueous phase oxidation (Met→MetO) and disulfide dimerization. Poor system cleanliness leads to a significant loss of activity, which is its most significant physicochemical weakness.

Poor BBB penetration raises questions about whether peripheral administration can achieve effective concentrations in the central nervous system. Intranasal and intraventricular delivery only yield impressive data in PD/AD animals; subcutaneous/peritoneal delivery is based on human imagination, not evidence.

All hard data are limited to cellular and rodent data; in humans, only observational data exist (low plasma levels in coronary heart disease/AD patients, high levels in centenarians). There are zero RCTs for the therapeutic use of humanin, which is not recognized by the FDA/EMA and falls under the RUO (Reactive Organic Drug) category.

The anti-apoptotic mechanism is a double-edged sword: Bax inhibition + STAT3 activation is theoretically possible in proliferative/malignant cell lines. Although animal chemotherapy models show no tumor protection, off-target effects in human tumor environments have been ruled out.

Research-grade commercially available formulations are mainly HNG, with wild-type HN being less common. A bottle labeled "Humanin 10mg" may actually contain HNG; sequence prediction requires checking the COA before making any decisions.

Storage Conditions:

The finished product is generally a sterile lyophilized peptide powder. The peptide chain is sensitive to oxidation, humidity, and repeated temperature fluctuations. Oxidative modifications will destroy its ability to bind to Bax and its receptor recognition ability, directly resulting in loss of biological activity.

Unopened lyophilized powder: Sealed, protected from light, and kept dry; store at -20°C. It is recommended to use within 12 months. Alternatively, store at -80°C, sealed, and protected from light; shelf life can reach 24 months. Repeated freeze-thaw cycles are strictly prohibited.

After reconstitution, the liquid sample must be aliquoted into single-use portions, stored at -80℃ in a light-proof, sealed container, and used within 6 months; only very short-term temporary storage is permitted at -20℃; long-term storage at 4℃ is prohibited, and storage at room temperature is strictly forbidden.

Opening operations should be performed quickly in a low-temperature, dry environment to minimize the powder's contact time with air, moisture, and oxygen; keep away from ultraviolet light and strong oxidizing agents.

Precautions

The RUO label means "Do Not Use on Humans." Any subcutaneous/nasal spray for personal use violates regulations; purchased vials are not equivalent to medicine.

Wild-type HN cell experiments should be performed at 1–100 nM, and HNG at 0.1–10 nM. Animal intraperitoneal/nasal assays should be designed at the μg/kg level; do not directly multiply cell concentration by body weight.

The COA must include HPLC ≥98%, MS identification, Met oxidation peak ratio, and endotoxin; for HN labeled but actually HNG, this must be clearly stated in the sequence field.

Caution regarding drug stacking: It can interfere with phenotypic interpretation when used with STAT3 inhibitors (such as static targeted therapies), Bax probes, and IGF-1 systems. A negative sequence should be included in the control group (e.g., a scramble using the PNC-29 approach).

Avoid during pregnancy and lactation, in cases of active solid tumors, during periods of active autoimmune disease, or in patients sensitive to mammalian cell sources; broad-spectrum anti-apoptosis activity poses a reverse risk in these scenarios.

Injection site pain/rare headaches are often self-reported noise in the community, but no research-grade safety profile has been established for humans. If persistent discomfort occurs, do not try to endure it.
