PNC-27 is a synthetic chimeric peptide, constructed from the HDM-2 binding fragment of the p53 tumor suppressor protein and a membrane-penetrating sequence. It is intended for research purposes only and has not yet been approved for human clinical application. Breaking away from traditional approaches that target intracellular signaling pathways, it utilizes the HDM-2 protein specifically expressed on the tumor cell membrane as a recognition anchor to selectively target malignant cells. It is a highly valuable molecular tool in the study of tumor cell lysis mechanisms and is widely used in in vitro and animal model studies of hematologic malignancies and solid tumors.
Molecular Essence and Design Origins: PNC-27 is neither a small molecule drug within the traditional chemotherapy framework nor an immune checkpoint inhibitor. It is a chimeric peptide with a precisely arranged 32 amino acid residues. One end of its molecular backbone carries the p53 protein HDM-2 binding domain (residues 12–26), while the other end tethers a membrane resident peptide (MRP) guide sequence. The initial concept for this structure stemmed from the classic approach of competitively blocking the nuclear binding of p53 to HDM-2, thereby prolonging the p53 half-life and restoring its transcriptional activation function. However, the experimental trajectory significantly deviated from the intended script-PNC-27 triggered a membrane lysis event on the cancer cell membrane surface completely independent of p53 activity, thus opening up an unprecedented cell death paradigm of "peptide-induced transmembrane pore formation" (Poptosis).
Core Advantage: A New Pathway Avoiding Drug Resistance
Its greatest advantage lies in the uniqueness of its mechanism. Because it disrupts the cell membrane through physical means, it perfectly avoids the drug resistance developed by cancer cells to traditional chemotherapy drugs or targeted drugs. In vitro and in vivo experiments have confirmed that it has extremely strong killing power against various malignant tumors such as breast cancer, pancreatic cancer, and acute myeloid leukemia, and has shown extremely high safety in animal models.
Storage Conditions
Lyophilized powder: Original bottle -20℃, sealed, protected from light, and sealed with desiccant, stable for 24 months; short-term storage at 2–8℃, do not exceed 6 months. Before opening, allow the bottle to reach room temperature to prevent condensation and hydrolysis. Reconstitute (sterile water/water containing 0.9% benzyl alcohol for antibacterial purposes/neutral PBS, slowly push along the wall without vortexing): Use within 7–28 days (manufacturer range varies, conservatively 7–14 days) at 2–8℃ protected from light. Aliquoting to smaller portions can extend storage time at -20℃, but repeated freeze-thaw cycles should be avoided. Discard any solution that is cloudy, colored, or contains flocculent material. Amphiphilic peptides readily adsorb onto the tube walls; use low-adsorption tubes and avoid using ordinary polypropylene for long-term storage of dilute solutions.
Precautions:
For research use only. Not for human or animal clinical administration. Not for use as a diagnostic or therapeutic agent. All operations are limited to laboratory in vitro experiments and compliant animal experiments.
Use sterile, enzyme-free water as the preferred reconstitution solvent to avoid heavy metals and impurities in ordinary buffer solutions that could accelerate peptide chain degradation. Complete powder opening quickly at low temperatures to minimize contact with air and moisture.
This peptide exhibits membrane perturbation properties. Protective gloves and goggles must be worn during experimental procedures to avoid direct skin and mucous membrane contact with the powder and reconstitution solution. In case of spillage, rinse thoroughly immediately.
Repeated freeze-thaw cycles and prolonged exposure to room temperature can cause peptide chain oxidation and conformational changes, directly weakening pore-forming bioactivity and causing fluctuations in experimental data. Strict control over storage and handling procedures is necessary.
Current conclusions are all based on in vitro and animal models. A complete human clinical trial has not yet been completed, and human therapeutic effects cannot be directly inferred. When citing literature, the research stage boundaries must be clearly indicated.
The HPLC purity of different batches can affect cell experiment results. Before formal experiments, it is recommended to use the activity parameters on the Certificate of Authorisation (COA) as a reference benchmark.
