Storage (lyophilized powder reagent diameter):
Unreconstituted lyophilized powder: Store at -20℃ in a sealed, light-protected, and dry place. Most brands indicate a shelf life of 1–2 years; at -80℃, it can last 2–3 years. It is hygroscopic, and Trp is susceptible to light oxidation. Use desiccant and wrap the bottle with aluminum foil; avoid frequent opening.
After reconstitution: Dissolve in sterile water/physiological saline/BAC at a concentration of 0.1–1 mg/mL; at -20℃, aliquots can be stored for approximately 1 month; at -80℃, approximately 6 months; at 4℃, it can only be stored for a few days to two or three weeks, not for long-term use.
Major Contraindications: Repeated freeze-thaw cycles, exposure to moisture, and UV exposure.
Precautions:
Before opening, allow the container to equilibrate at room temperature for 15–20 minutes. Otherwise, the negative pressure inside the bottle will draw in air and moisture, causing the nonapeptide to clump and directly reducing its activity.
Slowly add water along the wall, avoiding vortexing-the N-terminal Trp is already easily cleaved; further agitation due to shear force will reduce the in vitro half-life from 15 minutes to 5 minutes, resulting in distorted Western blotting/EEG results.
The fifth Asp must be in the α-configuration (α-Asp5); the β-Asp5 isomer is inactive. High temperatures, extreme pH levels, and prolonged solution storage can cause isomerization. If the COA (Copy of Account) doesn't specify configuration identification, be mindful of this before conducting functional experiments.
Low-concentration adsorption glass: Use low-adsorption tubes/polypropylene for preparing stock solutions whenever possible, avoid ordinary glass bottles. A concentration of a few μg/mL can absorb half a tube.
It's only a research reagent: Sigma's D4072 is labeled "for research, not for human use," meaning it's not a legal drug for subcutaneous injection. Even for in vivo experiments, ethical/IACUC (Intra-Aggregate Intake and Control) approval is required. Don't treat the supplier's label as a prescription.
When performing sleep EEG, remember to include an artificial cerebrospinal fluid infusion control-many early "induced delta wave" positive results were later found to be partly due to sleep pressure caused by the injection procedure itself.
