VIP 10mg/vial Is Used For Anti-inflammatory Treatment Of Respiratory Diseases. CAS: 40077-57-4

Vasoactive intestinal peptide (VIP) is a 28-amino acid neuropeptide belonging to the glucagon/secretin superfamily. It shares 68% amino acid sequence identity with pituitary adenylate cyclase activating polypeptide (PACAP). VIP functions as both a neurotransmitter and a neuroregulator in vivo, and is widely distributed in the central nervous system, digestive tract, and peripheral nervous system.

Product Introduction

Application Methods

1. Reconstitution Guidelines
VIP has excellent water solubility. It is recommended to reconstitute it using sterile water for injection, sterile PBS, or 1% dilute acetate buffer to improve complete solubility and initial stability. Add the solution slowly along the bottle wall, gently swirling at low speed. Vigorous vortexing, sonication, and high-temperature heating are prohibited to avoid peptide chain denaturation and adsorption inactivation. A clear, colorless solution is normal; the appearance of flocculation, turbidity, or precipitation indicates degradation and inactivation, and the solution should be discarded immediately.

Due to its extremely short half-life, it is recommended to prepare and use immediately, aliquoting only once, and avoiding repeated freeze-thaw cycles.

2. In Vitro Cell Experiments
Prepare sterile gradient working solutions for macrophage, airway epithelium, intestinal epithelium, nerve cells, and fibroblast models; primarily used for verifying inflammatory pathways, immune polarization, apoptosis, barrier function, and fibrosis mechanisms. It is recommended to aliquot the stock solution and store it at -80℃ to avoid rapid activity decay.

3. In vivo animal experiments
The mainstream routes of administration in scientific research are subcutaneous injection and intraperitoneal injection; due to their extremely rapid elimination from the body, animal experiments often use 1-2 doses daily to maintain effective concentrations. Nebulization/nasal drops can also be used for respiratory models, depending on the experimental protocol, strictly adhering to the model's dose gradient settings, and should be prepared and administered immediately.

Application Scenarios and Research Progress

1. Chronic Inflammation and Autoimmune Diseases

VIP exhibits broad anti-inflammatory and immunomodulatory effects in models of rheumatoid arthritis (RA), inflammatory bowel disease (IBD), and multiple sclerosis (MS):
* Inhibits inflammatory infiltration, pannus formation, and bone erosion in collagen-induced arthritis (CIA)
* Downregulates Th1 cytokines (IFN-γ) and upregulates Th2 cytokines (IL-4, IL-10)
* Inhibits matrix metalloproteinase-2 (MMP-2) activity

2. Chronic Inflammatory Response Syndrome (CIRS)

In the Shoemaker biotoxin-related disease protocol, VIP is used as the final step (Step 12), provided that all prior biomarkers (VCS, HLA-DR, TGF-β1, C4a, etc.) have returned to normal. Its role is to restore immune homeostasis, improve cerebral blood flow perfusion, and enhance cognitive function.

3. Respiratory Diseases

Pulmonary Arterial Hypertension (PAH): The synthetic form of Aviptadil lowers pulmonary artery pressure through pulmonary vasodilation and has entered Phase III clinical trials.

Sarcoidosis and Asthma: Inhalation administration is being explored, utilizing its dual bronchodilatory and anti-inflammatory effects.

COVID-19 Related ARDS: Aviptadil received FDA Fast Track designation, but subsequent Phase III trials failed to meet the primary endpoint, and it did not receive Emergency Use Authorization.

4. Gastrointestinal Diseases

Regulates intestinal smooth muscle motility, improving symptoms of irritable bowel syndrome (IBS) and inflammatory bowel disease (IBD).

Promotes intestinal mucosal epithelial repair, protecting the integrity of the intestinal barrier.

Stimulates mucus secretion, maintaining intestinal homeostasis.
