Precautions:

Slowly mix water along the wall, do not vortex-D(KLAKLAK)₂ is amphiphilic and acts like a surfactant; vortexing creates bubbles that are difficult to eliminate, and shear force promotes aggregation; slowly roll to mix. The solution should be clear and slightly yellow; discard if it is foggy or flocculent. Disulfide bonds are critical: For CoA (Chemical Oxide), use both non-reducing and reducing dual MS to verify cyclization rate. Avoid using it in the same tube as DTT/TCEP, and avoid prolonged alkaline storage. Low-concentration adsorption: For nM–μM working concentrations, avoid using low-adsorption polypropylene tubes and adding 0.1% BSA carrier; the actual amount in the wells can drop by half.

Four key parameters for CoA: HPLC ≥97–98%, MS confirmation ~2460/2611 Da (standard definition salt type), reducing/non-reducing dual mass spectrometry, and endotoxin testing. This peptide is more difficult to synthesize than linear peptides; cheaper alternatives often contain incompletely cyclized fragments.

 

RUO (Renal Oxide) and nephrotoxicity: The label should clearly state "Laboratory Research Use Only, not for human consumption." For in vivo assays (even in mice), IACUC (Intra-Anaerobic Microscopy) is essential, and monitoring BUN, creatinine, and urine sediment is crucial. Do not treat it as a regular metabolic peptide for long-term 4-week administration.
